Sintesis protein berlangsung di dalam inti sel dan di dalam ribosom. Masih ingatkah kamu tentang ribosom? Ribosom adalah organel sel yang ukurannya kecil dan padat serta berfungsi sebagai tempat untuk sintesis protein.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Namanya juga sintesis, pasti membutuhkan bahan baku. Nah<\/span><em><span style=\"font-weight: 400;\">,<\/span><\/em><span style=\"font-weight: 400;\"> bahan baku dalam sintesis protein adalah asam amino. Jumlah asam amino yang terlibat di dalam sintesis protein adalah 20. Namun, urutan asam amino tersebut dikode sepenuhnya oleh DNA. Transkripsi<\/strong><\/h3>\n<p><span style=\"font-weight: 400;\">Transkripsi adalah proses pencetakan RNA oleh DNA. RNA yang terbentuk dibedakan menjadi tiga, yaitu tRNA, mRNA, dan rRNA. Urutan asam amino akan ditentukan oleh basa nitrogen yang menempel di rantai mRNA. Tempat untuk menempelnya RNA polimerase pada DNA disebut sebagai promoter. Promoter berfungsi untuk menunjukkan tempat dimulainya transkripsi dan menentukan DNA cetakan dari dua rantai yang ada. Proses ini disebut sebagai inisiasi.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Dua rantai DNA yang terdiri dari rantai cetakan (<\/span><em><span style=\"font-weight: 400;\">sense<\/span><\/em><span style=\"font-weight: 400;\">) dan komplemen (<\/span><em><span style=\"font-weight: 400;\">antisense<\/span><\/em><span style=\"font-weight: 400;\">) mulai terpisah. Untuk promoter menempel di bagian rantai cetakan (<\/span><em><span style=\"font-weight: 400;\">sense<\/span><\/em><span style=\"font-weight: 400;\">).<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">RNA mulai dibentuk oleh RNA polimerase dari titik awal promoter dan bergerak terus di sepanjang rantai cetakan DNA. Akibatnya, heliks DNA terbuka secara berurutan sepanjang 10 \u2013 20 basa nitrogen. Nukleotida-nukleotida ini dirangkai dari arah 5\u2019 <\/span><span style=\"font-weight: 400;\">-&gt;<\/span><span style=\"font-weight: 400;\"> 3\u2019. Proses ini disebut sebagai elongasi.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Basa nitrogen yang dibentuk pada RNA merupakan komplemen basa nitrogen pada rantai DNA <\/span><em><span style=\"font-weight: 400;\">sense<\/span><\/em><span style=\"font-weight: 400;\">, yaitu sebagai berikut.<\/span>\n<ul style=\"list-style-type: circle;\">\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Basa T pada DNA untuk cetakan A pada RNA.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Basa C pada DNA untuk cetakan G pada RNA<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Basa A pada DNA untuk cetakan U pada RNA.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Basa G pada DNA untuk cetakan C pada RNA.<\/span><\/li>\n<\/ul>\n<\/li>\n<li><span style=\"font-weight: 400;\">Setelah RNA polimerase mentranskripsi DNA terminator, proses transkripsi akan berhenti. Proses ini disebut sebagai terminasi.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Kemudian, RNA akan lepas dari RNA polimerase. Hal itu mengakibatkan DNA heliks tertutup kembali.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">mRNA yang dihasilkan dari tahap transkripsi akan keluar menuju inti sel ke ribosom.<\/span><\/li>\n<\/ul>\n<h3><strong>2. Translasi<\/strong><\/h3>\n<p><span style=\"font-weight: 400;\">Translasi adalah penerjemahan kode genetik RNA menjadi urutan asam amino. Pada tahap ini, akan disintesis polipeptida menggunakan kode genetik dari mRNA di dalam ribosom oleh tRNA dengan bantuan enzim sintetase tRNA-aminoasil. Translasi juga terdiri dari tiga proses seperti transkripsi, yaitu inisiasi, elongasi translasi, dan terminasi translasi. Lalu, tRNA sebagai initiator yang memuat antikodon UAC akan membawa asam amino metionin. Asam amino tersebut akan melekat pada kodon AUG. Ingat, metionin merupakan asam amino awal dalam sintesis protein.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Selanjutnya, asam amino lain akan ditambahkan oleh enzim tRNA-aminoasil hingga dihasilkan rantai polipeptida lengkap.<\/span><\/li>\n<li style=\"font-weight: 400;\"><span style=\"font-weight: 400;\">Translasi semacam itu akan terus berlangsung sampai muncul <\/span><em><span style=\"font-weight: 400;\">kodon stop<\/span><\/em><span style=\"font-weight: 400;\">, yaitu UAA, UGA, atau UAG.
